ExaccType DPYD Kit
Qualitative detection of DPYD variants by multiplex qPCR

Product Overview
- Detects 7 predefined DPYD variants in a multiplex qPCR workflow
- Compatible with 1% EDTA whole blood and extracted genomic DNA
- 4 reactions per sample support efficient research workflows
- Detects the functional HapB3 variant c.1129-5923C>G directly
The ExaccType DPYD Kit is a Research Use Only multiplex real-time PCR assay for the qualitative detection of seven predefined DPYD variants from purified genomic DNA or 1% EDTA whole blood. Four multiplex reactions cover all included targets, while supplied positive and negative controls support run monitoring. The assay is designed for use on compatible real-time PCR instruments supporting FAM™, HEX™, ATTO®550 and ROX detection channels. The ExaccType DPYD Kit supports DPYD variant analysis in pharmacogenetics, molecular genetics, and translational research applications.

Scientific Background
DPYD Gene
DPYD encodes dihydropyrimidine dehydrogenase (DPD), an enzyme involved in pyrimidine metabolism. Genetic variation within DPYD is frequently investigated in pharmacogenetics, molecular genetics and translational research.
DPYD Variants in Research
Variants within the DPYD gene are commonly included in pharmacogenetics and biomarker research programmes. Molecular analysis of these variants supports studies of genetic variation, population genetics, assay development and investigations into metabolic pathways.
ExaccType DPYD Kit Concordance
The ExaccType DPYD Kit demonstrated concordant genotyping results when compared with alternative research genotyping methods. Genomic DNA extracted from 29 specimens with known DPYD variant status (characterized using NGS, microarray or qPCR-based methods), the Seraseq DPYD DNA Mutation Mix (SeraCare, Cat. No. 0750-9502), and one synthetic DNA sample for each variant were evaluated with the ExaccType DPYD Kit*.
*Analytical performance data were generated using the APIS DPYD HT qPCR Kit. The ExaccType DPYD Kit is the BIOTYPE-branded configuration of the assay.
**One of which was the Seraseq DPYD DNA mutation mix. One synthetic DNA material per variant was evaluated.
