Genomic DNA from whole blood is the workhorse material of clinical genetics, pharmacogenomics, hereditary disease testing, and HLA typing. Despite decades of refinement in manual extraction chemistry, variability introduced by manual pipetting, operator fatigue, and inconsistent incubation times remains a persistent source of result irreproducibility — particularly in high-throughput settings.

The IntelliPrep Blood Genomic DNA Extraction Kit delivers consistent, high-purity genomic DNA from 200 µL whole blood or buffy coat using the fully automated IntelliPrep TRIO 240 platform. The magnetic-pipette-based workflow handles every step from lysis through elution, removing manual touchpoints and delivering ready-to-use gDNA in approximately 60 minutes. Expected yields of 4–8 µg per 200 µL blood input meet the requirements of genotyping arrays, qPCR assays, WGS, and NGS panel sequencing.

Workflow

The extraction proceeds automatically in six stages: (1) Automatic sample loading from sample tube to extraction strip (2) Cell lysis using a proprietary buffer system with controlled heating, releasing genomic DNA while denaturing RNases and proteases. (3) Magnetic bead binding of released genomic DNA. (4) Multi-step washing to remove haemoglobin, lipids, cellular debris, and lysis buffer residuals — common inhibitors of downstream PCR. Final wash step ensuring high elution purity. (5) Air-dried bead drying step to ensure complete removal of residual wash buffer, preventing inhibitor carry-over into the gDNA eluate. (6) Elution of intact, high-molecular-weight genomic DNA into TRIO Elution Tubes.

The protocol is specially optimized for blood-derived specimens. DNA integrity is confirmed by gel electrophoresis, and functional quality is validated by downstream qPCR performance.
